Current study aimed to evaluate the antioxidant and antiproliferative effects of CN, extracted in chloroform, methanol, and water, on cancer cell lines. Antioxidant properties of CN were evaluated using DPPH, galvinoxyl, nitric oxide, and hydrogen peroxide based radical scavenging assays, whereas the tumoricidal effect was tested on HepG2, IMR32, NCL-H23, SNU-1, Hela, LS-174T, K562, Raji, and IMR32 cancer cells using MTT assay. Our data showed that CN in chloroform extract was a good antioxidant against DPPH and galvinoxyl radicals, but less effective in negating nitric oxide and hydrogen peroxide radicals. Chloroform extract exerted the highest antiproliferative effect on K-562 (%) and Raji cell lines (%) at 100 μg/ml and the other five cancer cell lines in a concentration-dependent manner, but not on IMR-32 cells. Fourteen known compounds were identified in chloroform extract, which was analysed by gas chromatography—mass spectra analysis. Recently, C. nutans has become popular for the treatment of cancer among Malaysian. Thus, the aim of this present study is to prove the anti cancer activity of C. nutans extracts in a treatment of cervical cancer as claimed by local people. Aqueous and methanol extracts were extracted from the leaves of C. nutans and phytochemical screening was performed for determination of secondary metabolites. The cytotoxic activities of both aqueous and methanol extracts were investigated against HeLa cell by using MTT assay. The mode of cell death was examined by Hoechst 33258 nuclear staining. The secondary metabolite constituents detected in C. nutans aqueous extract are terpenoids and flavonoids, whereas methanol extract contains terpernoids, alkaloids and flavonoids. Results also showed that C. nutans aqueous extract exerted a significant cytotoxic effect on HeLa cells (IC50=13±0.82 µg/ml) but no IC50 was detected by methanol extract. No significant cytotoxic activities (IC50 = not detected) were observed in normal kidney cell line, Vero, treated with both aqueous and methanol extracts of C. nutans which showed the cytoselective property of the extracts. However, HeLa and Vero cells treated with control drug, tamoxifen showed a significant cytotoxicity effects with IC50 values of 3.8±0.19 µg/ml and 2.2±0.029 µg/ml respectively. Hoechst 33258 stained showed the aqueous extract of C. nutans induced cell death on HeLa cells via apoptosis. In previous studies, the extraction of bioactive compounds from C. nutans by conventional Soxhlet solvent extraction has been described, but this method shows limitations in terms of selectivity, extraction yield and toxicity of the solvents employed. In this study, phytochemical compounds were extracted from leaves and stems of C. nutans by microwave-assisted extraction (MAE), pressurized microwave-assisted extraction (PMAE), supercritical carbon dioxide extraction (SFE) and Soxhlet method to investigate the best technique in terms of yield, extraction time and recovery of bioactive compounds: phenols, flavonoids, phytosterols and β-sitosterol. The extracted phytocompounds and phenolics were characterized by gas chromatography mass spectrometry (GC/MS) and ultra performance liquid chromatography (UPLC). The results showed that MAE was the best technique to achieve a high yield and a maximal total polyphenol content (11.30 ± 0.39 mg GAE/g DM) and flavonoids content (and 4.66 ± 0.20 mg GAE/g DM), whereas SFE was the best method for phytosterols and β-Sitosterol extraction. P-MAE merely enhanced the polyphenol and flavonoids yield to 14.56 ± 0.77 mg GAE/g DM and 5.29 ± 0.30 mg QE/g DM respectively, without significant variations on the type of compounds obtained. Lindau is a traditional medicinal plant belonging to the Acanthaceae family. Its therapeutic potentials have been increasingly documented particularly the antiviral activity against Herpes Simplex Virus (HSV), anti-cancer, anti-oxidant, anti-inflammatory and immunomodulatory activities. However, majority of these studies used crude or fractionated extracts and not much is known about individual compounds from these extracts and their biological activities. In the present study, we have isolated four compounds (CN1, CN2, CN3 and CN4) from the hexane fractions of C. nutans leaves. Using NMR spectroscopic analysis, these compounds were identified to be shaftoside (CN1), stigmasterol (CN2), β-sitosterol (CN3) and a triterpenoid lupeol (CN4). To determine the immunosuppressive potential of these compounds, their effects on mitogens induced T and B lymphocyte proliferation and the secretion of helper T cellcytokines were examined. Among the four compounds, stigmasterol (CN2) and β-sitosterol (CN3) were shown to readily inhibit T cell proliferation mediated by Concanavalin A (ConA). However, only β-sitosterol (CN3) and not stigmasterol (CN2) blocks the secretion of T helper 2 (Th2) cytokines (IL-4 and IL-10). Both compounds have no effect on the secretion of Th1 cytokines (IL-2 and IFN-γ), suggesting that β-sitosterol treatment selectively suppresses Th2 activity and promotes a Th1 bias. CN3 was also found to significantly reduce the proliferation of both T helper cells (CD4+ CD25+) and cytotoxic T cells (CD8+ CD25+) following T cell activation induced by ConA. However, the underlying molecular mechanisms for its action and whether C. nutans can offer protection on stroke damage in brain remain largely unknown. In the present study, we demonstrated protective effects of C. nutans extract to ameliorate neuronal apoptotic death in the oxygen-glucose deprivation model and to reduce infarction and mitigate functional deficits in the middle cerebral artery occlusion model, either administered before or after hypoxic/ischemic insult. Using pharmacological antagonist and siRNA knockdown approaches, we demonstrated ability for C. nutans extract to protect neurons and ameliorate ischemic injury through promoting the anti-apoptotic activity of peroxisome proliferator-activated receptor-gamma (PPAR-γ), a stress-induced transcription factor. Reporter and chromatin immunoprecipitation promoter analysis further revealed C. nutansextract to selectively increase CCAAT/enhancer binding protein (C/EBP)β binding to specific C/EBP binding site (-332~-325) on the PPAR-γ promoter to augment its transcription. In summary, we report a novel transcriptional activation involving C/EBPβ upregulation of PPAR-γ expression to suppress ischemic neuronal apoptosis and brain infarct. In this work, polyphenols have been recovered from C. nutans by microwave-assisted extraction (MAE). The influence of the main process parameters has been analyzed: ethanol concentration in the extraction solvent mixture, microwave energy applied and solvent-to-feed ratio. Model results have been correlated with mathematical models of the extraction process, that allow linking the observed variations in extraction curves with modifications of mass transfer and equilibrium parameters. Moreover, in order to assess the energy efficiency of the process, results were studied and explained in view of the specific energy absorbed during the microwave treatment (J/g). Results indicate that microwave pre-treatment improves extraction rate by a factor of 2–5, yielding the best results when a water–ethanol solvent mixture with 50%vol of ethanol is used at a solvent-to-feed ratio (S/F) of 14 mL/g. CNP-1-2 comprised approximately 87.25% carbohydrate and 9.37% protein. Monosaccharide analysis suggested that CNP-1-2 was composed of L-rhamnose, L-arabinose, D-mannose, D-glucose and D-galactose with a molar ratio of 1.30:1.00:2.56:4.95:5.09. Methylation analysis, FT-IR, and 1H NMR spectroscopy analysis revealed that CNP-1-2 might have a backbone consisting of 1,4-linked Glcp, 1,3-linked Glcp, 1,3-linked Manp, 1,4-linked Galp, 1,2,6-linked Galp and 1,2,6-linked Galp. Its side chain might be composed of 1-linked Araf, 1,6-linked Galp and 1-linked Rhap residues. Lindau extracts using two neutrophil-dependent acute inflammatory models and, in order to elucidate underlying cellular mechanisms, the effects of the extracts on human neutrophil responsiveness was investigated. Materials and methods The in vivo inflammatory models examined were carrageenan-induced paw oedema and ethyl phenylpropiolate-induced ear oedema in rats. Myeloperoxidase (MPO) activity was assayed as an indicator of neutrophil migration. Human neutrophil functional responsiveness was determined by measuring fMLP-induced chemotaxis, superoxide anion generation (SAG), and release of MPO and elastase. Apoptosis was assessed morphologically and flow-cytometrically. Neutrophil viability was assessed by trypan blue exclusion and MTT cytotoxicity assays. Results Both extracts induced powerful dose-dependent inhibitory effects in both edema models in rats. Importantly, there was a significant inhibition of MPO activity in the inflamed tissue indicating that the anti-inflammatory effect of the extracts is associated with reduced neutrophil migration. Although both extracts did not affect neutrophil viability or apoptosis, treatment of neutrophils with the extracts concentration-dependently inhibited fMLP-induced chemotaxis, SAG, and MPO and elastase release. Conclusions These findings suggest that the powerful anti-inflammatory properties of Barleria lupulinaLindl and Clinacanthus nutans (Burm.f.) This plant has diverse and potential medicinal uses in traditional herbal medicine for treating skin rashes, insects and snake bites, lesions caused by herpes simplex virus, diabetes, and gout in Malaysia, Indonesia, Thailand and China. Phytochemical investigations documented the varied contents of bioactive compounds from this plant namely flavonoids, glycosides, glycoglycerolipids, cerebrosides and monoacylmonogalatosylglycerol. The pharmacological experiment proved that various types of extracts and pure compounds from this species exhibited a broad range of biological properties such as anti-inflammatory, antiviral, antioxidant, and anti-diabetic activities. The findings of toxicity study showed that extracts from this plant did not show any toxicity thus it can be used as strong therapeutic agents for specific diseased conditions. However, further experiments on chemical components and their mode of action showing biological activities are required to elucidate the complete phytochemical profile and assess to confirm their suitability for future drugs. 憂遁草,是一種生長在靠近赤道的低海拔疏林中或灌叢內,潮濕砂質土壤裡的一種高大草本植物,成熟期一般為3--9個月。研究證實憂遁草的類黃酮物質含量極高,還有豐富的多種人體所必需的礦物質、維生素、胺基酸等,當然最令人在意的,還是它有一些神奇的功效,國內及國際學者都已在相關實驗中,逐一獲得證實!

 

      憂遁草主要分佈於馬來西亞、印尼爪哇、加里曼丹、中國雲南、廣西、廣東及海南等地,其中又以海南五指山熱帶雨林地區的「小尖葉憂遁草」品質最好,營養成分也最高。

「小尖葉憂遁草」跟大葉的憂遁草,品種是不同的,在栽培上也比較困難,不容易存活;以海南五指山來說,屬於低緯度地區,熱帶雨林氣候,全年降水豐富,多低海拔疏林和灌叢,土壤呈潮濕砂質,特別適宜小尖葉憂遁草的生長;當地政府鼓勵農民施作,生產基地也逐年擴大,在五指山山林地區,野生的憂遁草健康青綠、葉片小而尖細,被稱為“小尖葉憂遁草”,當地黎族中醫認為“小尖葉憂遁草”效果最好,也被稱為救命草。同時,小尖葉憂遁草也被國際公認為最好的憂遁草品種。

 

     「小尖葉憂遁草」,我們率先引進台灣,目前正在全力製成「忘憂茶」,將於近日與大家見面,但是,數量確實有限,由於小尖葉憂遁草營養成份極佳,複方的「忘憂茶」溫和調理,對健康維持全面且周到,應為養生首選;純葉使用有過寒之慮,不可不慎!
